A novel noninvasive method to detect rejection after heart transplantation BJMBR
Hu,Jun; Xie,Xin; Li,Yuan; Wang,Shuang; Feng,Qing; Wang,Xin; Liang,Daoyan.
Prompt and accurate detection of rejection prior to pathological changes after organ transplantation is vital for monitoring rejections. Although biopsy remains the current gold standard for rejection diagnosis, it is an invasive method and cannot be repeated daily. Thus, noninvasive monitoring methods are needed. In this study, by introducing an IL-2 neutralizing monoclonal antibody (IL-2 N-mAb) and immunosuppressants into the culture with the presence of specific stimulators and activated lymphocytes, an activated lymphocyte-specific assay (ALSA) system was established to detect the specific activated lymphocytes. This assay demonstrated that the suppression in the ALSA test was closely related to the existence of specific activated lymphocytes. The ALSA...

Anatomical Characteristics of Infra-Lamina Ridge in Chinese Population International Journal of Morphology
Li,Xiao-he; Cai,Yong-qiang; Wang,Xin; Gao,Shang; Li,Zhi-jun.
The aim of our study was to observe the occurrence rate and morphological characteristics of Infra-Lamina ridge (ILR) in Chinese population and provide a comprehensive reference for its clinical implication. Vertebrae columns of 82 sets of Chinese adult skeletons were collected. The shape of ILR was classified into three types; the occurrence of it in different sides and sexes were counted. The length, width, thickness of ILR were measured using a caliper with 0.02 mm accuracy. The ILR was usually found to occur from C7 to L5, with its frequency higher from T8 to 12, and highest at T10being up to 80.5%. There was significant statistical difference to the occurrence rate by side, sex and vertebral level. The largest values in its length and width were from...

Identification of microRNAs and mRNAs associated with multidrug resistance of human laryngeal cancer Hep-2 cells BJMBR
Yin,Wanzhong; Wang,Ping; Wang,Xin; Song,Wenzhi; Cui,Xiangyan; Yu,Hong; Zhu,Wei.
Multidrug resistance (MDR) poses a serious impediment to the success of chemotherapy for laryngeal cancer. To identify microRNAs and mRNAs associated with MDR of human laryngeal cancer Hep-2 cells, we developed a multidrug-resistant human laryngeal cancer subline, designated Hep-2/v, by exposing Hep-2 cells to stepwise increasing concentrations of vincristine (0.02-0.96'µM). Microarray assays were performed to compare the microRNA and mRNA expression profiles of Hep-2 and Hep-2/v cells. Compared to Hep-2 cells, Hep-2/v cells were more resistant to chemotherapy drugs (∼45-fold more resistant to vincristine, 5.1-fold more resistant to cisplatin, and 5.6-fold more resistant to 5-fluorouracil) and had a longer doubling time (42.33±1.76 vs 28.75±1.12'h,...

LncRNA HULC mediates radioresistance via autophagy in prostate cancer cells BJMBR
Chen,Changxuan; Wang,Kaizhen; Wang,Qian; Wang,Xin.
Prostate cancer (PCa) is the second leading cause of cancer death in men. Irradiation is one of the available options for treatment of PCa, however, approximately 10–45% of PCa are resistant to irradiation. We aimed to explore the role of long non-coding RNA highly upregulated in liver cancer (HULC) in the sensitivity of PCa cells to irradiation. Survival rate, cell apoptosis, cycle, expressions of related proteins, and caspase-3 activity were assessed to explore the effects of HULC on sensitivity of PCa cells to irradiation. Expression of HULC in DU-145, PC3, LNCaP, and RWPE-1 cells was determined and the influence of HULC on DU-145 cells was explored. Then, PC3 cells aberrantly expressing HULC were implanted into NOD-SCID mice for tumor xenograft study....

miR-26a-5p protects against myocardial ischemia/reperfusion injury by regulating the PTEN/PI3K/AKT signaling pathway BJMBR
Xing,Xiaowei; Guo,Shuang; Zhang,Guanghao; Liu,Yusheng; Bi,Shaojie; Wang,Xin; Lu,Qinghua.
Reperfusion strategies in acute myocardial infarction (AMI) can cause a series of additional clinical damage, defined as myocardial ischemia/reperfusion (I/R) injury, and thus there is a need for effective therapeutic methods to attenuate I/R injury. miR-26a-5p has been proven to be an essential regulator for biological processes in different cell types. Nevertheless, the role of miR-26a-5p in myocardial I/R injury has not yet been reported. We established an I/R injury model in vitro and in vivo. In vitro, we used cardiomyocytes to simulate I/R injury using hypoxia/reoxygenation (H/R) assay. In vivo, we used C57BL/6 mice to construct I/R injury model. The infarct area was examined by TTC staining. The level of miR-26a-5p and PTEN was determined by...

Properties of realgar bioleaching using an extremely acidophilic bacterium and its antitumor mechanism as an anticancer agent Biol. Res.
Chen,Peng; Xu,Ruixiang; Yan,Lei; Wu,Zhengrong; Wei,Yan; Zhao,Wenbin; Wang,Xin; Xie,Qinjian; Li,Hongyu.
Abstract Realgar is a naturally occurring arsenic sulfide (or Xionghuang, in Chinese). It contains over 90% tetra-arsenic tetrasulfide (As4S4). Currently, realgar has been confirmed the antitumor activities, both in vitro and in vivo, of realgar extracted using Acidithiobacillus ferrooxidans (A. ferrooxidans). Bioleaching, a new technology to greatly improve the use rate of arsenic extraction from realgar using bacteria, is a novel methodology that addressed a limitation of the traditional method for realgar preparation. The present systematic review reports on the research progress in realgar bioleaching and its antitumor mechanism as an anticancer agent. A total of 93 research articles that report on the biological activity of extracts from realgar using...
